If you want to see and do everything, then I'd say no. In my opinion, you need at least two days - minimum!!

we went in january /february
and we rode everyride except for 4 things (all of which go in circles)
dumbo-aladdin-carrousel-and the tomorrowland indy speedway
which doesn't go around in circles
and we even got to do buzz which was closed then -longest line in the MK that day
Around here, however, we don't look backwards for very long. We keep moving forward, opening up new doors and doing new things... and curiosity keeps leading us down new paths. - Walt Disney
With a good plan, yes, it is possible. It also depends on what time of year.
A day and half.
Assuming that not everything will appeal to you, then I think it can be done. Make some things a priority and go from there. Use fastpass when it looks to be the best plan. Be flexiable and enjoy what you can see and do rather than go postal over things you can't.

This year for Spring Break, there are a few days that with EMH the MK is open from 7 am till 3 am... you might have a chance of doing it all if you stayed for the eholw thing, otherwise I think you need at least 2 days, preferably 3....
Chuck Norris can.

Now, to answer the question...
IF you are well-rested going into the day, and IF you go at a time when crowds are minimal, and IF the EMH keeps it open for longer than 12 hours...yes. Otherwise, take two or three days to do it.
